Fig. 4

Overexpressing miR-181a-5p counteracts the attenuating influence of Gm37494 up-regulation on IL-1β-induced chondrocyte damage. Oe-Gm37494 and miR-181a-5p mimic were simultaneously transfected into 1L-1β-treated chondrocytes. A the expression of miR-181a-5p in 1L-1β-treated chondrocytes measured by qRT-PCR; B–C chondrocyte proliferation determined by CCK-8 (B) and EdU (C) assays; D the levels of the inflammatory factors TNF-α, IL-6, and IL-10 in 1L-1β-treated chondrocytes evaluated by ELISA; E chondrocyte apoptosis tested by flow cytometry; F the expression of apoptotic proteins Bax, Bcl-2, and cleaved caspase 3 in 1L-1β-treated chondrocytes detected by western blot. Data were expressed as mean ± standard deviation, and each experiment was repeated 3 times. *P < 0.05, compared with the oe-NC + mimic NC group; #P < 0.05, compared with the oe-Gm37494 + mimic NC group
