Sample processing overview. A total of 41 serum samples were collected at time of surgery from subjects with (n = 10) and without (n = 31) heterotopic ossification. Serum samples were pooled by disease state and subjected to an isobaric tag for relative and absolute quantitation (iTRAQ) mass spectrometry (MS) analysis. The iTRAQ data was used to drive the selection of specific proteins to target via a selected reaction monitoring (SRM) MS technique, a qualitative MS technique that enables the robust quantification of specific peptides within a single subject’s serum sample
